The Antibacterial Activity of Rhizophora mucronata leaves fractions against Staphylococcus aureus and the most active fraction FT-IR and GC-MS Profile

Abstract

Staphylococcus aureus is a microbial cause of infections worldwide. However, the widespread and irrational use of antibiotics has led to various cases of antibiotic resistance. Therefore, further research on natural substances has the potential to againts the S. aureus bacteria growth. The ethanol extract of R. mucronata leaves is classified as a strong antibacterial activity against S. aureus bacteria. However, the antibacterial activity from fractions R. mucronata leaves has never been reported. The objective of this study is to determine the antibacterial activity of fractions obtained from the Vacuum Liquid Chromatography (VLC) of ethanol extracts from R. mucronata leaves against Staphylococcus aureus and to identify their secondary metabolite profiles. The extraction method used sonication followed by separation using VLC with a gradient polarity solvent system. The activity of these fractions as antibacterial was determined by disk diffusion method. Samples were prepared at a single concentration of 25%. Antibacterial activity was measured based on the diameter of inhibition zones around the disks. The most active fraction was qualitatively identified for secondary metabolite profiles using TLC (Thin Layer Chromatography), Gass Chromatography Mass Spectroscopy (GCMS) and Fourier Transform Infrared (FTIR). The result revealed that fraction O was the largest diameter of inhibition zone (12.75 mm) classified as strong antibacterial activity. The secondary metabolites at fraction O were alkaloids, flavonoids, phenolics, tannins, and terpenoids. The FTIR spectra of fraction O showed the functional group at frequency 1639 cm-1, 3342 cm-1, and 1047 cm-1 identified the C=O, O-H, and C-O groups, respectively. The GC-MS analysis showed that in fraction O was identified 8 compounds such as Cyclohexane (0.98%), Cetyl glycidyl ether (1.33), palmitoleic acid (2.16%), Caryophyllene oxide (2.40%), Palmitic acid (3.33), D-Bornesitol (9.87%), 1-Eicosanol (19.56), and 1-hentetracontanol (totaling 60.38%). These compounds were reported has antimicrobial activity. So, the fraction O can develop as the antibacterial agent.
